細胞分化機構與訊息傳遞,基因選殖與表現,基因轉錄調控機構解析,本研究室可分成三大方向:

1.      細菌素機能與調控機構解析
細菌所生產的細菌素 (抗生物質一種 )只能殺死帶有近緣基因的細菌,而對其他親緣性較遠的物種細胞無任何的抗生效果。這種帶有特異性活性抗生物質,多數是由蛋白質所組成。近年來在針對細菌素相關研究中除了被應用在食物保存、 DNA/RNA等生物分子的修飾外、動植物病害防治外,更被發現在癌細胞的增殖或生存的抑制上有顯著的效果。本研究室主要是尋找新規的細菌素基因並加以選殖,這些新規的細菌素的殺菌活性部位、對近緣基因的細菌的辨識 (受體 )部位、細胞外進入細胞內的機構、更進一步的確認其反應機構及蛋白結構。此外,細菌素調控機構也需要進一步的研究。藉由細菌素的特性的了解,以期可以應用在醫學,工業,農業,分子生物學或各種生物防治上。

2.      細菌素胞外分泌機制解析
細菌細胞內物質分泌到細胞外需要透過分泌系統,針對分泌系統的組成不同可分別為第 I型、第 II型…等不同分泌系統,不同的分泌系統會則會分別分泌著不同的物質到細胞外,但個別分泌系統間的組成蛋白是互相獨立;我們在近年的研究中發現此一結果顯示 Erwinia屬的細菌素 Carocin S1 Carocin S2 蛋白的胞外輸送分泌機制同時與分屬 T3aSS T3bSS的第三型分泌系統的部分蛋白質有間接或直接的關係。本研究目的在於探討當輸送 Carocin S1 Carocin S2 蛋白至胞外時, T3aSS T3bSS分泌系統間的關係為何,其在 Erwinia屬中是否混合成一個新型態的蛋白質分泌系統以專責分泌低分子量細菌素。 Carocin S1 Carocin S2蛋白又是如何透過此一分泌系統被輸送至胞外,這三種不同系統的蛋白彼此間的關係及其作用機制是本研究的重點。

3.      人類細胞激素多型性表現與臨床疾病關係解析
當外來病源體進入身體後體內會立即產生多種細胞激素以刺激一連串的免疫反應,藉以此抵抗病源體對身體所產生的傷害。然而需多的疾病產生有可能是與細胞激素基因表現上發生變化,導致細胞激素的活性產生變化,並造成身體的免疫障礙,進而導致疾病的產生。本研究室與基隆長庚醫院胸腔內科合作針對敗血症、肺結核。。等國人在臨床上常見的疾病進行細胞激素多型性表現分析,以從而了解國人在細胞激素多型性表現與臨床疾病關係。
