背景

  1. 十字花科蔬菜黑腐病菌感染幾乎所有的十字花科蔬菜,造成全世界農業經濟上重大損失, 防治以施用化學農藥為主,但是會有葉片殘毒及環境污染的問題。我們想要找出菌之致病基因 (蛋白), 並且了解這些致病基因 (蛋白)的調控,以進一步設計非農藥防治策略。
  2. 南投縣仁愛鄉的一次痢疾感染事件及此事件後的四年中,自感染痢疾的病人分離出174痢疾桿菌菌株。 以脈衝式電泳將這些菌株進行分子分型,獲得一型的流行菌株 (五年內持續出現) 與十數型脈衝電泳圖形非常相似的非流行菌株 (五年內僅出現一或二次)。我們想要找出流行菌株持續出現的原因或非流行菌株僅出現一或二次的原因。
  3. 分枝桿菌已超過100 種,除了其中會引起肺結核的結核分枝桿菌外,其他總稱為非結核分枝桿菌。 非結核分枝桿菌有些為人體或環境中的非致病菌,有些為伺機性致病菌,有些為致病菌,引起人類肺部或局部的疾病。 由於分枝桿菌生長緩慢,生化鑑定繁瑣耗時,常延誤治療。我們想要發展分子鑑定的方法,可於環境及人類的檢體中, 快速鑑定出結核分枝桿菌及非結核分枝桿菌至種的階段。
  4. 非結核分枝桿菌近年來出現抗藥菌株,因此治療多採用二種以上的抗生素一起使用, 但是由於服藥期長,病人常有副作用。我們一方面想要篩選出更多的可用的抗生素,並且研究最佳的二種的抗生素組合, 以減少病人服藥的劑量及時間。另一方面也想要分離出非結核分枝桿菌的抗藥基因,研究抗藥菌株的抗藥機制。
